They can combine two opposite statements. While ''although'' is a subordinate conjunction which requires a subject and a verb, ''in spite of'' is a prepositional phrase which requires a noun, pronoun or gerund.

e.g. Although it rained, we enjoyed our picnic.

e.g. In spite of the rain, we enjoyed our picnic.
